Abstract Objective To investigate the effect of agmatine on hyperoxia-induced acute lung injury(HALI) in rats, and to find a theoretical basis for the drug treatment of HALI. Methods Forty-eight clean-grade SD rats were randomly divided into six groups (n=8), control group, hyperoxia group, agmatine A group, agmatine B group, agmatine C group and agmatine D group. Rats in control group were fed in air, hyperoxia group, agmatine A group, agmatine B group, agmatine C group and agmatine D group were fed in hyperoxia chamber. Blood gas was extracted from the arteries of rats 48 hours after modeling, and oxygen index (OI) and respiratory index (RI) were calculated. The left lower lung of rats was taken to measure the wet/dry mass ratio of lung in rats. The contents of TNF-α, IL-6 and IL-1 in alveolar lavage fluid and the activity of SOD and MDA were measured. Results ①Compared with the control group, OI and SOD in the hyperoxia group and the agmatine groups were significantly decreased, while W/D, RI, TNF-α, IL-6, IL-1 were significantly increased (all P<0.05); compared with the hyperoxia group, OI and SOD in the agmatine groups were significantly increased, while W/D, RI, TNF-α, IL-6, IL-1 were significantly decreased (all P<0.05). ②Compared with group A, group B and group C, with the increase of agmatine dosage, OI increased gradually and RI decreased gradually, but there was no significant difference between group C and group D (P> 0.05). ③The alveolar structure of the control group was normal, the alveolar wall of the hyperoxia group and the agmatine groups were broken and fused into bullae, the alveolar septum was obviously edema and widened, a large number of inflammatory cells infiltrated, edema fluid and inflammatory cells appeared in the alveolar cavity, and a small number of red blood cells were observed. With the increase of agmatine dosage, the damage of alveolar structure in rats of group B, group C and group D was gradually alleviated, while the changes of alveolar structure in rats of group C and group D were not significant. Conclusion Agmatine has a definite protective effect on HALI in rats, and the best protective dose may be 400 mg/kg.
