Protective effects of rhIL-11 for intestinal injury associated with sepsis |

Abstract Objective To evaluate the protective effect of recombinant human interleukin - 11(rhIL-11)on
intestinal injury associated with sepsis. Methods Eighty SD rats were divided into rhIL-11
treatment group(30 rats),sepsis group(30 rats),normal group(10 rats)and sham group(10 rats)
randomly. Septic rat model was established by cecal ligation and perforation(CLP). rhIL-11 treatment group and sham group were given subcutaneous injection of rhIL-11 500 μg/(kg·d)additionally
for 3days. However,rats of sepsis group or normal group were injected with balanced solution
(10 mL/ kg·d).Samples of intestine were collected at 24 h and 48 h after CLP to perform
histopathological study and molecular analysis. The mRNA expressions of Bax were evaluated by transcription-polymerase chain reaction. The protein expressions of JAK1 in lung were assayed by western
blot. Results ①The injury and the necrosis of small intestinal mucosal were reduced significantly
in treatment group compared with that of sepsis group. ②The expression level of Bax mRNA was lower in treatment group compared with that of sepsis group(P<0.05). ③The level of JAK1 protein
expression in the treatment group was observably higher than that of the sepsis,sham,and normal
groups(P<0.05). Conclusion rhIL-11 can reduce the intestinal injury associated with sepsis,the
main mechanism maybe rhIL-11 can regulate JAK/STAT signal pathway and reduce the expression of Bax.
