Effect of triterpenoids bardoxolone methyl on the oxidative stress signaling pathway of Nrf2-ARE/heme oxygenase-1 (HO-1) in rats with acute respiratory distress syndrome |

Abstract Objective To observe the effect of triterpenoids bardoxolone methyl (BARD) on the oxidative stress signaling pathway of Nrf2-ARE/heme oxygenase-1 (HO-1) in rats with acute respiratory distress syndrome (ARDS). Methods One hundred rats were divided into two groups using a random number table: BARD treatment group and saline treatment group. After feeding a week, two groups were divided into two subgroups according to whether the rats were established an ARDS model. A1 group: ARDS model+BARD group, A2 group, ARDS model + saline group, B1 group: normal group+BARD group, B2 group: normal group + saline group. The animals continued feeding for 72 hours and all survived mice were sacrificed to obtain specimens. Histopathology changes of lung tissues were observed under light microscope. The expression of Nrf2 and HO-1 proteins in lung tissue were detected by immunohistochemistry. RT-PCR method was employed to detect the expression of Nrf2 mRNA and HO-1 mRNA in lung tissue. Results Pathological structure of lung tissue group A2 was the most serious and the pathological score was the highest; Group A1 was less damaged than group A2; and group B1 and group B2 were generally normal. Expression of Nrf2, HO-1 protein: Nrf2 and HO-1 protein in groups A1(3.78±0.37,2.86±0.16) and groups B1(3.01±0.34,2.47±0.19) were higher than those in groups A2(1.82±0.31, 2.01±0.21)and B2(0.92±0.13,1.41±0.17); group A1 were higher than group B1; and group A2 were higher than group B2(Pall<0.05). Expression of Nrf2 mRNA, HO-1 mRNA: Nrf2 mRNA, HO-1 mRNA were most obvious in group A1(2.16±0.17, 1.45±0.12); group B1(1.79±0.14, 1.18±0.05) was higher than group B2(0.42±0.06, 0.68±0.11); and group A2(1.18±0.19, 1.01±0.07) were higher than group B2(0.42±0.06, 0.68±0.11,Pall<0.05). Conclusion Triterpenoids bardoxolone methyl can significantly increase the expression of Nrf2-ARE/HO-1 signaling pathway-related proteins in lung tissue, thus exerting antioxidant stress and reducing lung tissue damage during ARDS.
