The role of IL-33 in a murine model of renal ischemia and reperfusion injury |

Abstract Objective To explore the role of IL-33 in a murine model of renal ischemia and re⁃ perfusion injury. Methods Forty-five male C57BL/6 mice were divided into three groups: sham group (sham); ischemia and reperfusion injury group(PBS + I/R); IL- 33 + ischemia and reperfusion injury group(rIL-33+I/R). In sham mice,an abdominal incision was made,followed by dissection of the bilat⁃ eral renal pedicle,with no further cross-clamping of arteries. PBS+I/R mice were administered(i. g. ) phosphate buffered solution 1 hour before surgery whereas rIL-33 + I/R mice were administered rIL-33 5 μg 1 hour before surgery. Mice in NS+I/R and rIL-33 + I/R groups were subjected to renal ischemia to induce acute kidney injury after 45-min clamping of bilateral renal arteries. Serum levels of creatinine and blood urea nitrogen levels were measured. H&E staining was undertaken to detect damaged renal tis⁃ sue. Myeloperoxidase(MPO)activity and immunofluorescent detection of IL-33 expression was under⁃taken for each group. Level of IL-17A was monitored by ELISA. Results The expression of rIL-33 in murine renal was increased during renal ischemia and reperfusion injury. Serum creatinine and blood urea nitrogen levels were elevated in PBS+ I/R and rIL-33+I/R groups compared with sham mice(P< 0.05),whereas those in the PBS+I/R group were significantly higher compared with the rIL- 33 + I/R group(P<0.05). The damage of renal tissue and neutrophils infiltration in rIL- 33 + I/R groups were significantly lower than in PBS+I/R mice. The MPO activity and level of IL-17A in rIL-33+I/R was re⁃ duced than that in PBS+I/R groups(P<0.05). Conclusion IL-33 can inhibit renal I/R injury by in⁃ hibiting release of IL-17A and neutrophils infiltration.
