Effect of curcumin pretreatment on heat stroke rat′s proteins related to intestinal mucosa and serum inflammatory factors in dry-heat environment |

Abstract Objective To investigate the effects of curcumin pretreatment on the expressions of HSP70, HMGB1 and ICAM-1 in heat stroke rat′s intestinal mucosa and serum inflammatory factors level in dry-heat environment. Methods 30 SD rats were randomly divided into normal control group (NC group), dry heat group (DH group) and curcumin pretreatment group (Cur group), with 10 rats in each group. NC and DH groups were gavaged with saline. Cur group was gavaged with 200 mg/kg curcumin, once a day for 7 days. After the end of gavage, DH and Cur groups were placed in the experimental cabin of the simulated dry-heat environment, temperature of (41±0.5)℃, relative humidity of (10±1)%, exposed 150 min to cause severe heatstroke model. The expression of HSP70, HMGB1 and ICAM-1 protein were detected by Western blotting in intestinal mucosa, the concentration of LPS, TNF-α, IL-1β and IL-6 were detected by ELISA method. Results The expressions of HSP70, HMGB1, ICAM-1 and the concentration of LPS, TNF-α, IL-1β and IL-6 significantly increased in DH group campared with NC group (P<0.05). The expressions of HSP70 significantly increased in Cur group (8.75±0.46) compared with DH group (2.17±0.14),P<0.05. The expressions of HMGB1, ICAM-1 significantly decreased in Cur group (6.95±0.42, 9.31±0.76) compared with DH group (17.76±1.54, 19.39±1.64),P<0.05, the concentration of LPS, TNF-α, IL-1β, IL-6 significantly decreased in Cur group compared with DH group. Conclusion Curcumin pretreatment may protect intestinal mucosa and reduce the migration of bacteria or toxins in intestinal cavity by regulating the expression of HSP70, HMGB1 and ICAM-1, thereby alleviating systemic inflammatory response caused by heatstroke in dry-heat environment.
